A bicistronic vector backbone for rapid seamless cloning and chimerization of αβT-cell receptor sequences.

To facilitate preclinical testing of T-cell receptors (TCRs) derived from tumor-reactive T-cell clones it is necessary to develop convenient and rapid cloning strategies for the generation of TCR expression constructs. Herein, we describe a pDONR™221 vector backbone allowing to generate Gateway™ compatible entry clones encoding optimized bicistronic αβTCR constructs. It harbors P2A-linked TCR constant regions and head-to-head-oriented recognition sites of the Type IIS restriction enzymes BsmBI and BsaI for seamless cloning of the TCRα and TCRβ V(D)J regions, respectively. Large-scale effects of migration and conflict in pre-agricultural groups: Insights from a dynamic model.

The debate on the causes of conflict in human societies has deep roots. In particular, the extent of conflict in hunter-gatherer groups remains unclear. Some authors suggest that large-scale violence only arose with the spreading of agriculture and the building of complex societies. To shed light on this issue, we developed a model based on operatorial techniques simulating population-resource dynamics within a two-dimensional lattice, with humans and natural resources interacting in each cell of the lattice. Extracellular vesicles (EVs) that are derived from stem cells are proving to be promising therapeutic options. We herein investigate the therapeutic potential of EVs that have been derived from different stem cell sources, bone-marrow (MSC) and human liver (HLSC), on mesangial cells (MCs) exposed to hyperglycaemia. By expressing a dominant negative STAT5 construct (ΔNSTAT5) in HG-cultured MCs, we have demonstrated that miR-21 expression is under the control of STAT5, which translates into Transforming Growth Factor beta (TGFβ) expression and collagen production.
